Extract of Pomegranate inhibition of advanced glycation

Progression and maturation were delayed significantly with extract of Punica granatum treatment. Photographs of fundus shows marked thickness of retina and dilated blood vessels in diabetic rats compared to control. Gliclazide shows mild increase in retinal thickness and no dilated blood vessels. Normal thickness of fundus and blood vessels was observed with extract of Punica granatum treatment.

RESEACH:

The study was planned to investigate the effect of extract of Punica granatum on the role of inhibition of advanced glycation end products in streptozotocin induced diabetic retinopathy. Albino Wistar rats were induced for diabetes by streptozotocin and divided into five groups as vehicle control (group I), disease control (group II), standard gliclazide (Group III), group IV, and V received 50, 100 mg/kg of extract of Punica granatum respectively, for a period of 12 weeks.

Blood glucose levels were estimated and Cataract progression due to hyperglycemia was monitored by slit lamp biomicroscope and fundoscope at the end of every 4 weeks, the biochemical parameters involved in the pathogenesis of cataract such as protein content, SOD, CAT, reduced glutathione (GSH), aldose reductase (AR), protein carbonyl content (PCO) and advanced glycation end products (AGE) were measured in the lens homogenate. Slit lamp observations indicated cataract due to hyperglycemia.

The treatment with extract of Punica granatum exerted improvement in antioxidant enzymes as SOD, CAT, GSH and inhibiting AR levels, and inhibition of PCO and AGE levels in lens homogenate. These results suggest that the extract of Punica granatum is effective against STZ induced cataract and is beneficial in inhibition of formation of AGEs in the progression of diabetic retinopathy.
